Production of alkaline amylase by Bacillus sp. under solid state fermentation (SSF) was optimized. The effect of wheat bran and rice bran were examined. The appropriate Incubation time, Temperature, pH, Inoculum size, Moisture content, Carbon source, Nitrogen source, Substrate concentration, Surfactant and Mutagenic effects were determined. Maximum yield of enzyme was achieved by employing wheat bran as substrate ((94±2) U/g) at 72hrs with 1:1.5 moisture content and pH-8. Inoculum size was found to be 30% for wheat bran. The increased growth of bacterium of mutagenic activity is seen with acrylamide solution.
