The study was aimed to find out the impact of glucose on the expression of deleted in colon cancer (DCC) and Wilson tumour (WT1), tumour marker genes, in liver cancer cells and normal kidney cells. Liver cancer cell lines HepG2, Hep3B, HCCM, Huh 7, PLC5, and normal kidney cell lines MDCK and HEK 293 were grown in a medium containing two different concentration of glucose. Crystal violet assay was performed to determine the colony forming ability of the liver cancer cells and it was found to be decreased in cells grown in lower concentration of glucose. Expression level of DCC and WT1 was determined by RT-PCR with specific primer for the genes from the total RNA of liver and kidney cells grown in media with lower and higher glucose concentration. Our results showed that the cells grown in lower concentration of glucose showed reduced expression of the marker genes as compared with the cells grown in higher concentration of glucose. Furthermore, this study identified that glucose plays a vital role in the growth of some of these liver cancer cells and normal kidney cells. Interrupting the supply of glucose in cancer cells may be an appealing therapeutic strategy for cancer.
